Implementation of high productivity perfusion cell culture at-scale in an integrated continuous manufacturing platform
Continuous manufacturing as a means of producing biologics is gaining momentum in the industry, as it allows for improved productivity, more consistent product quality attributes over a long-duration cultivation, and more efficient resource utilization. Just – Evotec Biologics is a technology company built from inception on a continuous manufacturing platform. This presentation demonstrates integrated continuous manufacturing development at the 3L-scale through 500L/1000L GMP-scale manufacturing with a focus on leveraging high cell density cultures and longer culture duration for achieving high productivity. The data presented will include key technology enablers such as expression vectors and hosts, perfusion, media concentrates, process automation and control, and challenges encountered with implementation both in process development and at GMP manufacturing scale. We will address challenges such as gas transfer, foaming, perfusion filters, custom-designed single-use consumables and equipment. Additionally, we will review data that highlights the integration of downstream enabling technologies developed at the 50L-scale and implemented in manufacturing to meet Just – Evotec Biologics’ goals for end-to-end continuous commercial biomanufacturing.
